Bone marrow mononuclear cells versus mesenchymal stem cells from adipose tissue on bone healing in an Old World primate: can this be extrapolated to humans? Arq. Bras. Med. Vet. Zootec.
Branco,E.; Miranda,C.M.F.C.; Lima,A.R.; Silva,K.S.M.; Cabral,R.M.; Miranda,M.S.; Ohashi,O.M.; Oliveira,E.H.C.; Silva,L.S.C.; Freitas,D.M.M.; Miglino,M.A..
ABSTRACT In veterinary medicine, the cell therapy is still unexplored and there are many unanswered questions that researchers tend to extrapolate to humans in an attempt to treat certain injuries. Investigating this subject in nonhuman primates turns out to be an unparalleled opportunity to better understand the dynamics of stem cells against some diseases. Thus, we aimed to compare the efficiency of bone marrow mononuclear cells (BMMCs) and mesenchymal stem cells (MSCs) from adipose tissue of Chlorocebus aethiops in induced bone injury. Ten animals were used, male adults subjected, to bone injury the iliac crests. The MSCs were isolated by and cultured. In an autologous manner, the BMMCs were infused in the right iliac crest, and MSCs from adipose tissue...

Cell therapy in dilated cardiomyopathy: from animal models to clinical trials BJMBR
del Corsso,C.; Campos de Carvalho,A.C..
Dilated cardiomyopathy can be the end-stage form and common denominator of several cardiac disorders of known cause, such as hypertensive, ischemic, diabetic and Chagasic diseases. However, some individuals have clinical findings, such as an increase in ventricular chamber size and impaired contractility (classical manifestations of dilated cardiomyopathy) even in the absence of a diagnosed primary disease. In these patients, dilated cardiomyopathy is classified as idiopathic since its etiology is obscure. Nevertheless, regardless of all of the advances in medical, pharmacological and surgical procedures, the fate of patients with dilated cardiomyopathy (of idiopathic or of any other known cause) is linked to arrhythmic episodes, severe congestive heart...

ABSTRACT: This study aimed to evaluate two protocols (PA and PB) that are used to obtain canine platelet-rich plasma (PRP) for cellularity. Twenty healthy dogs were used. Blood samples were collected and placed in two tubes of 3.2% sodium citrate. PA used double centrifugation at 210 x g, and 370 x g and PB used double centrifugation with 140 x g and 330 x g. The PRP samples from the protocols were examined in terms of their platelet, erythrocyte, and leukocyte count in the Neubauer chamber, differential leukocyte count and platelet morphological observation in blood smears. Data (mean and standard deviation) were analyzed with the 95% probability t-test (P <0.05) using Pearson’s correlation to test the relationship between platelets and erythrocytes,...

Effect of freezing on rabbit cultured chondrocytes Arq. Bras. Med. Vet. Zootec.
Filgueiras,R.R; Del Carlo,R.J; Alves,N.P; Viloria,M.I.V; McManus,C.M; Castro,M.B; Filgueiras,F.P.F; Monteiro,B.S; Paula,A.C; Farias,A.
This work evaluated the effect of freezing on chondrocytes maintained in culture, aiming the establishment of a cell bank for future application as heterologous implant. Chondrocytes extracted from joint cartilage of nine healthy New Zealand White rabbits were cultivated and frozen with the cryoprotector 5% dimethylsulfoxide for six months. Phenotypic and scanning electron microscopy analyses were carried out to identify morphological and functional differences between fresh and thawed cells. After enzymatic digestion, a total of 4.8x10(5)cells per rabbit were obtained. Fresh chondrocytes showed a high mitotic rate and abundant matrix was present up to 60 days of culture. Loss of phenotypic stability was notable in the thawed chondrocytes, with a low...

Human umbilical cord blood cells in infarcted rats BJMBR
Pinho-Ribeiro,V.; Maia,A.C.V.; Werneck-de-Castro,J.P S.; Oliveira,P.F.; Goldenberg,R.C.S.; Campos de Carvalho,A.C..
Therapy with bone marrow-derived cells has been used in ischemic patients with reported success. The aim of this study was to determine the therapeutic efficacy of fresh and frozen human umbilical cord blood cells (hUCB) in Wistar rats submitted to permanent occlusion of the left coronary artery. Three hours after myocardial infarction, 2 x 10(7) hUCB cells or vehicle were administered by intramyocardial injection. The animals were divided into five groups: control (N = 10), sham operated (N = 10), infarcted that received vehicle (N = 9), infarcted treated with cryopreserved hUCB (N = 7), and infarcted treated with fresh hUCB (N = 5). Cardiac function was evaluated by electrocardiogram (ECG) and echocardiogram (ECHO) before cell therapy, and by ECG, ECHO,...

Senni, Karim; Pereira, Jessica; Gueniche, Farida; Delbarre-ladrat, Christine; Sinquin, Corinne; Ratiskol, Jacqueline; Godeau, Gaston; Fischer, Anne-marie; Helley, Dominique; Colliec-jouault, Sylvia.
The therapeutic potential of natural bioactive compounds such as polysaccharides, especially glycosaminoglycans, is now well documented, and this activity combined with natural biodiversity will allow the development of a new generation of therapeutics. Advances in our understanding of the biosynthesis, structure and function of complex glycans from mammalian origin have shown the crucial role of this class of molecules to modulate disease processes and the importance of a deeper knowledge of structure-activity relationships. Marine environment offers a tremendous biodiversity and original polysaccharides have been discovered presenting a great chemical diversity that is largely species specific. The study of the biological properties of the...

Sonographic abnormalities in augmented bladder using porcine intestinal submucosa (SIS) Arq. Bras. Med. Vet. Zootec.
Rossetto,V.J.V.; Souza,P.M.; Doiche,D.P.; Mamprim,M.J.; Padovani,C.R.; Brandão,C.V.S..
ABSTRACT Among the different materials for bladder augmentation, porcine intestinal submucosa (SIS) is the most widely investigated and stands out for its ability as a cell scaffold. In this context, the ultrasound examination allows the detection of changes from the surgical procedure, enabling the early verification of potential complications and evaluation of patient outcomes. The aim of this paper is to describe the main sonographic findings in dogs submitted to cystoplasty using acellular SIS and seeded with homologous smooth muscle cells at 30 (M30) and 60 (M60) days postoperatively. Sonographic changes included irregularities and thickening of bladder wall especially at M30. Additionally, were visualized urinary sediment and uroliths in animals...
